Lowercase stays lowercase (often used for masked regions). IUPAC ambiguity codes are complemented too: RโY, KโM, BโV, DโH; S, W and N stay the same.

The complement pairs each base (AโT, CโG) in the same order, so it reads 3โฒ โ 5โฒ. The reverse complement also reverses it, giving the other strand in the standard 5โฒ โ 3โฒ direction, which is what primers and databases use.
If your sequence is the coding (sense) strand, the mRNA has the same sequence with U instead of T: use DNA โ RNA. If it's the template (antisense) strand, use RNA from template, which takes the reverse complement and writes U for T.
